Prolonged Illness best fits a disease-token Assassin deck, especially Nuu or Arakni builds that repeatedly create Bloodrot Pox, Frailty, and Inertia tokens and can force them to be destroyed during the same turn; however, it is mainly a blue pitch card and provides no immediate pressure, so it is weaker in aggressive lists. Cards such as Infectious Host, Plague Hive, and other efficient attacks or effects that create multiple disease tokens are generally better because they advance your game plan immediately rather than waiting for tokens to be destroyed, while defensive blues may also be preferable when you need consistency. Prolonged Illness can be worthwhile as a one- or two-copy value tool in a slower Nuu control or fatigue deck, but it should not usually see play in competitive Assassin lists unless your build reliably destroys several differently named disease tokens every turn.
